Théoriquement, le jus de pamplemousse peut augmenter les concentrations plasmatiques de la loratadine comme il le fait à d'autres médicaments qui sont des substrats du CYP450 3A4 enzymatiques de la voie. Le mécanisme proposé est l'inhibition du CYP450 3A4 médiée par le métabolisme de premier passage dans la paroi intestinale par certains composés présents dans les raisins. La signification clinique de cette interaction potentielle est inconnue. Les interactions signalées avec de puissants inhibiteurs de cytochrome p450 3A4 comme la clarithromycine, l'érythromycine et le kétoconazole ont produit des augmentations substantielles de l'aire sous la courbe concentration plasmatique-temps (ASC) de la loratadine et de son métabolite actif, descarboethoxyloratadine, sans associées à des changements dans le profil de sécurité du médicament.
